Vitamin K2 binds 17beta-hydroxysteroid dehydrogenase 4 and modulates estrogen metabolism

Vitamin K is a cofactor for gamma-glutamyl carboxylase, an enzyme that is important for blood coagulation. Recent studies have shown that vitamin K has other roles, in addition to post-transcriptional modification, such as bone metabolism and antitumoral actions; these findings have indicated that there might be unknown intracellular binding proteins that are specific for vitamin K. In this study, vitamin K-binding proteins were characterized by pull-down experiment using a chemically synthesized biotynylated vitamin K followed by mass spectrometric identification of the pull-downed components. The results indicated that 17beta hydroxy steroid dehydrogenase 4, apolipoportein E, and 40S ribosomal proteins S7 and S13 might be the candidates of the vitamin K-binding proteins. Subsequent experiments showed that vitamin K2 binds 17beta hydroxysteroid dehydrogenase 4 and decreases the intracellular estradiol:estrone ratio, which resulted in the inhibition of the amount of estrogen receptor alpha-binding to its target DNA. These results suggest a possible novel role for vitamin K in modulating estrogen function.
I've done a lot of research on Vitamin K2 but somehow overlooked this one.
Just a few short years, we now know that Vitamin K2 helps the regression of coronary atherosclerotic plaque, builds bone significantly and helps prevent tooth decay.
On a related note here's an article on teeth: http://nourishedmagazine.com.au/blog/articles/forgotten-tooth-decay-cure-heal-your-cavities-and-prevent-root-canals

wow! so K2 can help bring down estrodial e1 e2 levels if they are too high?

Thanks for this CS! I have high estradiol and take 25mcg vit-k2 that's in with my D-3. Unfortunately this study doesn't indicate what dosage would be effective.
This brings me to a related question about calcium and mineral intake....
Other than a men's multi, the only other mineral I supp with is Mag (400-600mg/day). My question lately is if I should add any calcium or potassium. I am wondering of I am throwing the balance off by only consuming mag?
This is the 3rd best selling mag on iherb. It is mag and potassium aspartate with taurine:
http://www.iherb.com/Now-Foods-Magnesium-Potassium-Aspartate-with-Taurine-120-Capsules/701?at=0
I almost ordered this last night but decided to stick with the mag citrate that I have had no problems with thus far.

Gromit137 - The link did not come up properly, is this one one you were asking about?
http://www.iherb.com/Life-Extension-Super-K-with-Advanced-K2-Complex-90-Softgels/14619?at=0
gonzalito - DIM is very safe, however there isn't enough research on K2 to compare them in terms of estrogen regulation. Both are anti-cancer, both inhibit MMP's (matrix metalloproteinases enzymes). K2 is important to regulate calcium homeostasis, lowers blood sugar and is more powerful than Co-Enzyme Q10, and it helpful against wrinkles. As part of my regimen I take a little K2 everyday, that's much cheaper than the life extension product.
Hopefully more research will come out because I think we've only seen the tip of the iceberg on what K2 can do.

Yep ... that's the one. Think one per day is enough?
Thanks.

Re: Vitamin K2 binds 17beta-hydroxysteroid dehydrogenase 4 and modulates estrogen metabolism
Any news concerning the dosage of vitamin K for lowering estradiol?...couldn't find any. I'm taking the recommended dose from Decalcify for the moment.
Probably the best foods for K vits are natto and chlorrella:
We found that even small quantities (10 g) of natto greatly increased by 225% (from 0.57±0.18 ng/ml to 1.28±0.46 ng/ml) vitamin K2 blood concentrations (MK-7: menaquinone-7) for a period of over 48 hours. Also, vitamin K1 concentrations increased by 152% (from 0.46±0.28 ng/ml to 0.70±0.21 ng/ml) four hours after the intake of chlorella. ...unfortunately, finding natto in Europe is not easy.
Will raising T3 work for lowering lowering estrogen and increasing progesterone (according to Ray Peat/dannyroddy, but can't find the quote right now)?

Iodine will help raise T3 and iodine also helps keep estrogen at bay.
