Curcumin attenuates Wnt/ß-catenin pathway, while necessary for hair growth? Empty Curcumin attenuates Wnt/ß-catenin pathway, while necessary for hair growth?

Please excuse me if I've misunderstood these articles...
But if I understand them correctly, isn't taking curcumin actually in some way contradictionary when trying to get hair (re)growth?

Natural derivatives of curcumin attenuate the Wnt/β-catenin pathway through down-regulation of the transcriptional coactivator p300

Transient activation of β-catenin signaling in cutaneous keratinocytes is sufficient to trigger the active growth phase of the hair cycle in mice
http://www.ncbi.nlm.nih.gov/pmc/articles/PMC196062/
"Our data lend strong support to the notion that a Wnt/β-catenin signal operating on hair follicle precursor cells serves as a crucial proximal signal for the telogen–anagen transition."

(...)

"Wnt signaling appears to be involved in patterning in the skin, as mice with a skin-specific deletion of β-catenin (Huelsken et al. 2001) or mice with constitutive overexpression of an inhibitor of Wnt signaling in skin (Andl et al. 2002) fail to develop hair follicles."

longhairedredhead - Over the years on this forum there have been many similar questions to this.

The short answer is that various proteins do not always have a good or a bad expression in all tissue. Moreover, their expressions are co-dependent on other factors.

For example, with respect to the Wnt/β-catenin pathway, this can be increased in cancer--especially colon cancer. So it is not unusual to find that natural substances, such as curcumin will attenuate Wnt/β-catenin pathway in these circumstances.

Curcumin will not have universal effects in all tissue or under all physiological conditions. This is by the way, why drugs are bad, because unlike curcumin or other natural substances, which act as biological response modifiers, a drug will just turn on or turn off something (by breaking an enzyme or blocking a physiological pathway). There have been hair loss drugs, that will turn on a protein that is good for hair, but also cause cancer in the process.

Another example of something natural that affects the Wnt signaling pathway is folate. Components of the Wnt signaling pathway become altered under folate-deficient conditions. The Wnt signaling pathway has multiple functions, affecting cell growth, differentiation, and development.

Thanks, CS!

So what it basically does, is attenuating expressions when they're elevated, while leaving already 'balanced' levels unaffected?

That's correct. For example, MMP-2 can degrade the extracellular matrix, making room for tumors to develop. So let's say in the case of one cancer (and there are many others), curcumin is useful as a treatment of highly metastatic melanoma cells by inhibiting matrixmetalloproteinase-2 activity.

This doesn't mean that curcumin will block MMP-2 under all conditions.

Hmm, I see you were already writing a reply at the time I edited my post.
For other folks to understand what this is about: I initially asked a question about curcumin and its MMP-2 inhibiting/attenuating properties, while MMP-2 is a 'hair growth promoting enzyme' according to a study cited here:

Thanks for your time clarifying this to me, CS! Smile
